Abstract: Somaclonal variation can be a useful tool for plant genetic improvement. In this study,the immature embryo of three wheat cultivars( lines) were used to induce variation via tissue culture,and regenerated somaclonal plants were characterized and selected in field. A total of 12 elite somaclonal variants were finally obtained according to their comprehensive agronomic traits in R5 generation. Also,the inheritance of stripe rust resistance of somaclonal line 4-8with resistance to stripe rust,which reported previously was determined and the F1 and F2genetic populations were constructed by crossing susceptible variety Mingxian 169 with line 4-8. The resistance to stripe rust was identified by inoculating Puccinia striiformis f. sp tritici at seedling stage in greenhouse and at adult stage in field,showed this line conferred excellent resistance to wheat stripe rust,especially immune to Tiaozhong 33( CY33),the main epidemic stripe race at present in Gansu; the results of inheritance analysis indicated that resistance gene in line 4-8 was controlled by a pair of dominant genes. The results provide evendence for developing of wheat new germplasm with resistance to stripe rust and breeding of new variety by using somaclonal variation.
