Optimization of liquid fermentation medium for Bacillus B13

Abstract: Objective To construct the culturing technology for the development of microecological preparation of Bacillus B13. Methods Four factors including brown sugar,wheat bran,soybean powder and urea were chosen through single factor experiment; three-level L9(34) orthogonal tests were performed with each factor. Results The optimal carbon source of the medium was glucose,and the optimal nitrogen source was soybean powder. The optimum medium composition was consisted of 1% of brown sugar,1% of wheat bran,0. 5% of soybean meal,1% of urea,0. 5% of K2HPO4,and 1,000 mL of water,with pH at 7.0-7.2. Conclusion The result of viable count of Bacillus based on the optimal medium was 7. 8 × 109 CFU /mL.
