HPLC-ESI-TOF /MS in Rapid Separation and Identification of Multiple Chemical Constituents in Leonuri Herba

Abstract: Objective: To explore the feasibility of rapidly separating and identifying the chemical components in Leonuri Herba by high-performance-liquid chromatography-time of flight mass spectrometry( HPLC-TOF /MS). Methods: Extract of Leonuri Herba as the test solution,rutin and wogonin solution as standard substances. Agilent poroshell 120 SB-C18 column(3 mm ×100 mm,2.7 μm) was adopted. The mobile phase was composed of acetonitrile-0.1% formic acid with a nonlinear gradient elution. The flow rate was 0. 4 ml /min. The UV detection was set at 254 nm and colum temperature was 25℃. 10 μl aliquot of residues were injected into the HPLC-ESI-TOF-MS system. Electron spray ionization and positive mode was adopted,the flow and temperature of carrier gas( N2) were 10 L/min and 350℃,respectively. The capillary voltage was 4 kV,the bombardment voltage was 150 V,the spectra were recorded in the range of 100 ~ 1 100 m /z. Results: A total of 7 kinds of chemical constituents were identified from Leonuri Herba by HPLC-TOF /MS in one run. The major constituents in Leonuri Herba were identified by time of flight mass spectrometry and structure-relevant fragment ions. Conclusion: A rapid and efficient HPLC-TOF-MS approach for identifying the chemical constituents of Leonuri Herba has been successfully established,which paves a way for the pharmacodynamic material basis and quality control of Leonuri Herba.
