Abstract: Objective:Pfs25 is the key antigen of tansmission-blocking vaccines.Nevertheless,the phase I clinical trails revealed that Pfs25 elicited a low immunity.Chemical conjugation of Pfs25 can improve its immunogenicity.We investigated the chimeric expression of Pfs25 in this study.Methods:Isocaudarner was used to construct recombinant plasmids(Pfs25)3/pPICZαA(pGAPZaA),then it was transformed to P.pastrosis PDI /GS115,X-33 by electroporation to obtain recombinant P.pastrosis strain.The positive yeast was expressed on flask scale,and the target protein(Pfs25)3 was preliminarily purified.Results:The expression level of three copies of the gene was low as detected by ELISA and SDS-PAGE.The purity of target protein was 83%.Conclusion:Three copies of Pfs25 gene have been expressed in Pichia pastoris,providing an experimental basis for the development of malaria vaccines.
