Abstract: The stem tips(1-2cm in length)from germ-free seedlings of Penstemon barbatus(Can.)Roth were used as explants to establish a propagation system through bud induction,proliferation and rooting.The stem tips cultured on MS medium containing 1.0mg/L 6-BA+0.05mg/L NAA for 30dgave an adventitious bud induction rate of 100% and a propagation coefficient of 9.8.The adventitious buds were subcultured every other 30d.The multiplication coefficient averaged 6.5in generations 1to 4,and the cluster buds grew well.The adventitious buds were trained on 1/2MS medium containing 0.2mg/L IBA+0.2mg/L NAA for 20d,and their rooting rate was as high as 100%.Their root system was well developed and the test-tube plantlets grew vigorously.One hundred and five rooted plantlets were then transplanted to matrix soil,and 98of them(91.42%)survived after 15days’culture.This protocol may be useful for micropropagation and genetic transformation studies of P.barbatus.
