Effects of allicin on the expression of connexin43 isoforms in hyperlipemic rat’s cardiac muscle

Abstract: OBJECTIVE To find the molecular mechanism of decreasing blood fat effect of allicin on hyperlidemic rat,we studied the expression of connexin43 in the myocardium before and after using the allicin. METHODS 50 Wistar rats were randomly divided into 5 groups: control group,model group,allicin group of high dose,middle dose and low dose( 100,50,25 mg·kg-1·d-1). Each group had 10 rats. Hyperlidemic rat model was established firstly,the blood which was obtained via vena caudalis and the indexes of TC,TG,LDL,HDL and NEFA in the serum were measured,the myocardial total RNA was extracted by Trizol method,to compare the expression of connexin43 in the following groups,hyperlipemia,normal and drug,we used the technique of RT-PCR,immunostaining and microconfoul,Western blot. RESULTS The concentrations of TC,TG,LDL and NEFA in serum increased( P < 0. 05),while that of HDL decreased( P < 0. 05). After treating with allicin,the concentrations of the preceding four indexes was cut down and the concentrations of the post one went up to nearly normal level. The mRNA expressions and protein expression of connexin43 in hyperlipemia group was decreased( P < 0. 05),while that of the drug group was enhanced( P < 0. 05). We could see the same result in immunostaining. CONCLUSION Hyperlidemic rat model is successfully established,allicin has the effect of lowering blood lipid. Furthermore, the molecular mechanism of allicin is connected with the change of connexin43 closely.
