Molecular Detection of Allelic Variations of Genes for Lipoxygenase and Polyphenol Oxidase in 104 Gansu Wheat Cultivars

Abstract: Flour color and appearance color of flour products are the important indicators for evaluating wheat quality.The activities of wheat lipoxygenase(LOX)and polyphenol oxidase(PPO)have relatively important effects on the flour whiteness and color of flour products.In this study,104wheat varieties released in Gansu province were used as experimental materials to detect the allelic variation of lipoxygenase gene TaLox-B1 and polyphenol oxidase gene Ppo-A1 and Ppo-D1 by using function marker LOX16,LOX18,PPO18,PPO16 and PPO29,in order to further investigate the distribution characteristics of the three genes in wheat varieties from Gansu.The results showed that,the fre-quencies of cultivars with genes of TaLox-B1aand TaLox-B1b were 22.12%and 77.88%,respectively;the distribution frequency of LOX high activity gene TaLox-B1ain winter wheat varieties from Gansu(30.56%)was higher than that in spring wheat(3.13%).The frequencies of cultivars with genes of Ppo-A1a,Ppo-A1b,Ppo-D1aand Ppo-D1b were 49.04%,50.96%,50.96% and 49.04%, respectively.Frequencies of allelic combinations of PPO genes for Ppo-A1a/Ppo-D1b,Ppo-A1a/ Ppo-D1a,Ppo-A1b/Ppo-D1band Ppo-A1b/Ppo-D1awere 28.85%,20.19%,20.19%and 30.77%, respectively.Wheat cultivars recently released in Gansu province had a relatively high proportion of LOX low-activity gene TaLox-B1b.and the genes of low PPO activity were dominant.Among those varieties,32varieties contained the desired allelic variation for LOX and PPO genes.
