Effects of Scorpion Purified Liquid on the Release of NO,TFPI,TF Induced by Thrombin in Cultured Vascular Endothelial Cells

Abstract: Objective To observe the effect of sc orpion purified liquid(SPL)on the release of nitric oxide(NO),tissue fact or pathway inhibit or(TFPI),tissue fact or(TF)induced by thrombin in cultured vascular endothelial cells(VEC).Methods The human umbilical vein endothelial cells(HUVECs)were cultured in vitro.The well grown 2to 3generation HUVECs were given different treatment respectively.The supernatant fluid were taken to measure NO,TFPI,The freezing and thawing liquid were taken to measure the activity of TF.Results Compared with the blank control group,thrombin group could obviously inhibit the release of TFPI on HUVEC(P<0.01).Compared with thrombin group,SPL in each dose group could antagonize the decrease of TFPI(P<0.05).Thrombin promoteed the express of TF on VEC,it was significantly lower in the control group(P<0.01).Compared with the thrombin group,SPL in each dose group was lower(P<0.01).Compared with the blank control group and thrombin group,the concentration of NO from sc orpion purified liquid each dose group was remarkably increased(P<0.01).Conclusion SPL could inhibit the release of TF,can antagonize the inhibition of the release of TFPI from HUVEC induced by thrombin and promote the release of NO from HUVEC induced by thrombin,suggesting that sc orpion protect the injured VEC induced by thrombin.
