Effects of Shenfu Injection on Primary Cultured Cardiocytes following Hypoxia/reoxygenation Injury in Neonatal rats

Abstract: Objective To investigate the protective effect of Shenfu injection(SFI)on primary cultured cardiocytes following hypoxia/reoxygenation(H/R)injury in neonatal rats and its possible mechanism.Methods The model of hypoxia/reoxygenation injury was developed in primary cultured cardiocytes of neonatal rats.The cultured cells were randomly divided into four groups:Group C(control)without any treatment;Group H/R with 1-hour hypoxia followed by 4-hour reoxygenation;Group SFI-L(1ow-dose SFI plus H/R),cardiocytes were pretreated with a low dose(50μmol/mL)of SFI f or 30min followed by H/R;Group SFI-H(high-dose SFI plus H/R),cardiocytes were pretreated with a high dose(100μmol/mL)of SFI f or 30min followed by H/R.The release of CK-MB and cTn-I in culture medium were evaluated.The content of malondialdehyde(MDA),activity of superoxide dismutase(SOD),and the apoptotic rate of cardiocytes were determined.Results The release of CK-MB and cTn-I in supematant,the content of MDA and the apoptotic rate of cardiocytes were significantly increased and SOD activity in cardiocytes was significantly decreased in group H/R(P<0.01vs group C),and they were markedly reduced in group SFI-L and SFI-H(P<0.01vs group H/R)except that SOD activity was markedly enhanced.Conclusion SFI could protect the primary cultured cardiocytes from hypoxia/reoxygenation injury by attenuating the lipid peroxidation reaction and inhabiting cardiocytes apoptosis.
