Abstract: 目的探讨同型半胱氨酸(HCY)对高糖培养的血管内皮细胞产生活性氧和细胞间黏附分子-1(ICAM-1)mRNA表达的影响。方法体外培养人血管内皮细胞(ECV-304),实验性模拟糖尿病的高HCY状态,随机分为4组:正常对照组、高糖(HG)组、同型半胱氨酸(HCY)组和高糖加同型半胱氨酸(HG+HCY)组。分别在培养12h、24h、48h进行相关的实验和检测。应用比色法检测培养细胞上清液中超氧化物歧化酶(SOD)活力和丙二醛(MDA)水平。用RT-PCR法测定(ICAM-1)mRNA表达的影响。结果与正常对照组相比,HG组和HCY组SOD活力明显降低(P<0.01),而MDA含量明显升高(P<0.01),ICAM-1mRNA表达上调且这种改变在HG组和HCY组更明显(P<0.01),并随时间延长有明显增加的趋势。结论 HCY可能通过氧化应激机制上调ICAM-1的表达,从而损伤内皮细胞,加重糖尿病血管病变的发生、发展。
