Abstract: The absorption,distribution and excretion of estriol labeled with tritium werestudied in female rats given a large dose of the steroid.The rate of excretion afteroral administration was compared with that after intramuscular injection.Apreliminary analysis of the properties of urinary radioactive metabolite was made.Following the intravenous injection of ~3H-estriol the radioactivity in the blooddeclined exponentially as a result of the rapid distribution of the labeled steroidto various tissues of the body.The estimation of radioactivity in tissues after in-tramuscular administration showed that much more radioactivity was accumulatedin liver than in other tissues and also remained there for a longer time.The plau-sible reasons for these results were discussed.The excretion of radioactivity viastool and urine in the first 3 days amounted to 70.9 % of the administrated dosegiven per os.while only 43.7% of the dose was excreted when given intramus-cularly indicating that orally administered steroid was more readily excreted.The urinary radioactive metabolites were found to be mainly the more polar estriolconjugates by TLC analysis.On the thirteenth day after drug administrationtrace amount of radioactivity was still present both in tissues and in stool and urine,suggesting that after a large dose,estriol or their metabolite may remain in thebody for a long time.
