Abstract: In mice the analgesic action was determined by means of the Haffner's method and modified Grewal's method. The intracerebral injection of morphine 50μg/kg produced marked analgesic action, lasting about 56 minutes. This dose was corresponding approximately to 1/100 of subcutaneous effective dose. When the dose was raised to 100 or 200/μg/kg, the analgesic action was more pronounced. It is thus suggested that the analgesic action of morphine is mainly a direct action on the brain centers. In 90 mice subcutaneous injection of morphine was administered daily till the tolerance developed, then morphine was given intracerebrally. It was demonstrated that the analgesic action of 100-200μg/kg by intracerebral route in the tolerant mice was decreased significantly (P0.01)in comparison with that in the nor-tolerant mice. After the tolerance disappeared, the same doses of morphine by intracerebral injection produced anew marked analgesia in mice. Hence. it is apparent that the tolerance to morphine occurs in the central nervous system. The intracerebral injections of corydalis B(dl-tetrahydropalmatine) 0.25-3 mg/kg had no analgesic action in mice.
