Abstract: In present study, in vitro matured and fertilized ovine oocytes were cultured long term or transferred into the recipients at 2~4-cell and morula—blastocyst stage, The development rate and conception rate were examined, respectively. The ovine oocytes were collected from local slaughterhouse and cultured in CO_2incubator(39℃, 5%Co_2in air) with TCM 199 containing 10% NSS(or FCS),hCG and estrogen for 24 to 28 pours. The matured oocytes with expanded cumulus were inseminated by fresh semen capacitated with Ca—ionophore A23187. After 7 to 10 hours, they were transferrd into development culture medium, i. e. Hepes buffered TCM 199 supplymented with 10% NSS (or FCS) and Pyruvate—Na. Part of 2~4—cell or morula—blastocyst stage embryos were transferred into recipients surgically and other embryos were cultured for 7 to 12 days for further examination of development of cleaved ova. The results showed that the cleavage rate after 48 to 72 hours of in emination in NSS treatment was 52.4%(182/347), significantly higher than that of in FCS treatment. 61 embryos of IVF were transferred into 20 recipinets, 10 of them were pregnant and 11 lambs were born. The conception rate reached 50%. 64.7%(312/482) of continuously cultured cleaved ova developed to morula—blastocyst stage (including hatched blastocysts).
