Abstract: In the present study, muscimol (a GABA receptor agonist), bicuculline (aGABA receptor antagonist), nipecotic acid (an inhibitor of GABA uptake) and3-mercaptopropionic acid (an inhibitor of GABA synthesis) were injected intothe dorsal raphe nucleus of rats with chronically embedded cannula, the dose ofeach drug was 4μg/μl. The influences of such injection on pain threshold andanalgesic effect of acupuncture (AEA) were investigated. Injection of muscimol or nipecotic acid caused respectively a rise in painthreshold (+20%-+40%, P0.05 and P0.01) and a marked enhancementof AEA. These effect was found to be apparent for at least 2h. The pain thresholdwas insignificantly changed after injection of bicuculline or 3-mercaptopropionicacid (P0.05), but injection of either drug could cause a significant decline ofAEA (P0. 05 and P0. 01). These results indicated that GABA in dorsal raphenucleus played an important role in modulation of pain transmission.
