Abstract: 目的:探讨雷帕霉素对大鼠梗阻性黄疸肝脏保护作用的相关研究.方法:采用胆总管结扎方法建立大鼠梗阻性黄疸模型,将54只SD大鼠随机分为假手术组(A)组18只,梗阻性黄疸(B)18只,梗阻性黄疸+STAT抑制剂雷帕霉素RPM处理组(C)18只.分别于术后1、3、5d处死大鼠,显色基质法测定各组内毒素含量,取肝脏组织测定超氧化物歧化酶(SOD)活性和丙二醛(MDA)含量,RT-PCR测肿瘤坏死因子基因表达.结果:B和C组TNF-α mRNA表达水平均增加,B组较C组内毒素水平含量低,肿瘤坏死因子基因表达水平低,SOD活性相对高,MDA含量低,有显著差异.结论:梗阻性黄疸时TNF-α表达明显升高,通过雷帕霉素干预,肝组织TNF-α的表达下降,SOD活性增加,MDA含量减少,可防止梗阻性黄疸所致失控性炎症反应性肝损伤.
