Abstract: 目的:研究葛根素对异丙肾上腺素(ISO)诱导的大鼠心肌组织中转化生长因子β1(TGF-β1)和血浆血管紧张素Ⅱ(Ang-Ⅱ)的影响.方法:采用ISO[5 mg/(kg·d)×14 d]皮下注射诱导大鼠心肌纤维化,给予葛根素100 mg/(kg· d)腹腔注射或卡托普利100 mg/(kg·d)灌胃,实验共8周.32只雄性SD大鼠随机分成4组:模型组8只;葛根素组8只;卡托普利组8只;葛根素+卡托普利组8只.实验末检测大鼠左室射血分数及缩短分数,计算左心室质量指数,取左室心肌组织进行VG染色,分别测算胶原容积积分,取左室游离壁心肌组织检测羟脯氨酸浓度,RT-PCR检测TGF-β1 mRNA水平和Western Blot测定TGF-β1蛋白,测定血浆Ang-Ⅱ浓度.结果:与模型组比较,葛根素和卡托普利均改善大鼠左室收缩功能(LVEF%,FS%,P< 0.01),减少左心室质量指数(P<0.01)及羟脯氨酸浓度(P<0.01),减少左室心肌组织TGF-β1(P< 0.05),降低血浆Ang-Ⅱ浓度(P<0.01).以葛根素+卡托普利最显著(P<0.01).结论:葛根素改善左心室收缩功能,抑制左室心肌组织中TGF-β1表达和降低血浆Ang-Ⅱ,葛根素与卡托普利合用作用更强.
