Studies on optimization of cellulase immobilization with amino-silane modified magnetic nanoparticles

Abstract: 以表面氨基功能化纳米磁性复合微球为载体,戊二醛为交联剂,对纤维素酶的固定化条件进行研究。先通过单因素试验确定了温度、时间、pH值、交联剂戊二醛浓度和酶浓度等对固定化酶活性的影响,进而根据中心因子复合原则设计pH值、温度和酶浓度3个因素响应面试验。对试验结果进行二次回归分析,得到了响应面模型,预测最优固定化条件:温度为21.1℃,pH为4.2,酶浓度为0.076,该优化条件下,固定化酶活有所提高,能较好地与响应面模型预测值相吻合。
