Abstract: 目的探讨宫内缺氧对新生大鼠海马CA3区血管内皮细胞生长因子(VEGF)表达与胶质细胞分化的影响以及当归的调控作用。方法选择雌性SD大鼠分别与雄性大鼠合笼。孕第14天健康SD雌性大鼠15只随机分为对照组、缺氧组和当归干预组,每组各5只。自孕第14天开始将当归干预组与缺氧组孕鼠置于低张氧三气培养箱中,制作胎鼠宫内缺氧模型,此前1h按8mL/kg分别予250g/L当归注射液和9g/L盐水尾静脉注射,对照组不予缺氧处理。3组孕鼠分娩当日每窝随机选取新生大鼠4只,取其脑组织多聚甲醛固定,石蜡包埋切片,胶质纤维酸性蛋白(GFAP)mRNA、VEGFmRNA原位杂交,400倍拍照。结果缺氧组新生大鼠海马CA3区GFAPmRNA与VEGFmRNA阳性细胞的积分吸光度(A)值均较对照组增大(Pa<0.05);当归干预组GFAPmRNA的A值较缺氧组减小(P<0.05),VEGFmRNA的A值较缺氧组增大(P<0.05)。结论一定时间、一定氧体积分数的宫内缺氧可刺激新生大鼠海马CA3区胶质细胞的增生,其机制可能与缺氧后导致新生大鼠海马CA3区VEGFmRNA表达增高有关;当归注射液可减弱宫内缺氧新生大鼠神经胶质细胞的增生,可能是通过上调VEGFmRNA的表达而改善缺氧环境所致。
