Abstract: 目的:研究5Gy γ射线放射损伤复合15%Ⅲ度烧伤小鼠骨髓基质细胞血小板生成素(TPO)基因表达的影响.方法:采用骨髓基质细胞培养、逆转录聚合酶链式反应、斑点杂交和原位杂交技术检测基质细胞TPO mRNA.结果:发现各伤组动物股骨骨髓巨核细胞数和外周血血小板数分别在伤后第7、13天和第3、7天降到最低值.骨髓基质细胞TPO mRNA量于伤后第3、13天出现明显的组间差异,表现在放射组和烧伤组明显低于正常水平,而复合伤组则明显高于正常值.两个时相点的变化趋势一致.结论:放烧复合伤后骨髓基质细胞分泌TPO的活性较单纯放射损伤和单纯烧伤活跃.
