Abstract: 目的:研究一氧化氮(NO)抑制单核-内皮细胞粘附的机制.方法:以硝普钠作为NO的供体,用细胞粘附实验、流式细胞仪荧光强度测定、细胞ELISA、Northern blot等方法分别从细胞、蛋白表达、基因表达方面研究NO对内皮细胞粘附分子(vascular cell adhesion molecule, VCAM-1)表达的影响.结果:NO抑制IL-1α诱导的单核-内皮细胞粘附,抑制IL-1α诱发的VCAM-1的表达并呈时间依赖性及浓度依赖性,NO在粘附分子VCAM-1的基因表达水平上亦呈现抑制作用.结论:NO抑制VCAM-1的表达是其抑制单核-内皮细胞粘附及抗动脉粥样硬化的机制之一.
