Abstract: 目的 探讨1-甲基,4-苯基,1、2、3、6-四氢砒啶(MPTP)对C57BL小鼠纹状体尾壳核内神经元结构及谷氨酸受体(GluR1)表达的影响.方法 通过MPTP注射至C57BL成年小鼠腹腔内建立帕金森病(Parkinson'sdisease,PD)动物模型.采用免疫细胞化学技术、显微图像分析技术检测小鼠纹状体尾壳核内谷氨酸受体GluR1的表达;应用透射电镜技术观察小鼠纹状体尾壳核内神经元超微结构的改变.结果 透射电镜观察实验组小鼠纹状体尾壳核神经元内线粒体肿胀,嵴结构部分模糊,部分内质网扩张,细胞核皱缩变形;GluR1的表达与对照组相比明显减少(p<0.05).结论 MPTP引起小鼠纹状体尾壳核神经元超微结构改变,其神经毒性作用可能与降低GluR1的表达有关.
