EFFECT OF STRESSFUL STIMULATIONS ON HYPOTHALAMIC VASOPRESSIN mRNA LEVEL OF RATS

Abstract: 实验在给予慢性应激刺激的Sprague Dawlay雄性大鼠中进行,用核酸斑点杂交技术观察大鼠下丘脑组织中血管升压素(AVP)mRNA水平变化,用异羟基洋地黄毒甙(DIG)标记的26个碱基长寡聚核苷酸作为检测探针.结果观察到:在给予电击足底结合噪声的应激刺激之后,尾动脉收缩压逐渐升高,到刺激第5天,刺激组与对照组动物之间尾动脉收缩压差别有统计学显著意义;到刺激第9天,刺激组尾动脉收缩压达最高值;以后各天,血压持续维持在高水平.给予动物电击足底结合噪声的应激刺激4 d以后,下丘脑组织中AVP-mRNA水平与对照组水平无显著差异;而给予刺激6 d或15 d之后,刺激组动物下丘脑组织中AVP-mRNA水平较对照组动物有显著升高(6 d:P<0.005;15 d:P<0.001).实验中还观察到,实验组与对照组动物之间血浆渗透压无显著差异.上述结果提示:电刺激足底结合噪声的应激刺激在引起大鼠血压升高的同时,还能使下丘脑组织中AVP-mRNA水平升高;AVP-mRNA水平升高的机制有待进一步的研究.
