Abstract: Objective:Study the anti-tumor effects of asiatic acid(AA)combined with chemotherapeutics cyclophosphamide(CTX) or 5-fluorouracil(5-Fu).Methods: The cytotoxic effects of the combination of AA and 5-FU on tumor cell lines HepG2 were detected by MTT assay.Mice transplanted with Heps solid tumor and ascites carcinoma were used to study the combination anti-tumor effect of AA(50,100 mg/kg) and CTX(20 mg/kg). Results: When AA(10,15,20,25 μmol/L) and 5-Fu(50,7.5,1 μmol/L) was combinated to treat the cells,the growth of HepG2 cells were inhibited obviously.On the other hand,AA prolonged the survival time of mice with Heps ascites carcinoma compared with those in CTX group and showed significant anti-tumor activity simultaneously.The protection of AA on hepatic GSH level insulted by CTX in mice with Heps solid tumor treated with AA and CTX confirmed the attenuated toxicity effects of AA,which might be associated with hepatopretection of AA. Conclusion: The present data suggest that AA attenuate the toxicity of some chemotherapeutics and may be helpful in cancer treatment with the combination of chemotherapeutics.
