Actions on mitochondria of carcinoma by different polar drugs

Abstract: OBJECTIVE:To investigate the mechanism how the polarity of drugs affect the function of mitochondria of lung cancer cells.METHODS:The lung cancer cells A549 were cultured and treated by sodium selenite, all-trans retinotic acid, sodium selenite combined with all-trans retinotic acid at the concentration of 20.0 μmol/L at certain time. The MTT colorimetric assay was used to analyze the proliferation inhibition rate. Flow cytometry (FCM) was used to detect the percentages of cells. A tansmission electron microscope was used to observe the changes of mitochondria. RESULTS:The proliferation inhibition rates of sodium selenite, all-trans retinoic acid group and combination group of the two drugs that had treated over 24-96 h were significantly higher than that of the 0.095% volume ethyl alcohol control group (P<0.01 or P<0.05), the differences of sodium selenite group and all-trans retinotic acid group were significant, P<0.05. The percentages of S phase cells of the three treated groups were significantly higher than those of the two controls, P<0.05. Contrast to it, G2/M phase was significantly lower, P<0.05, but it is not so significantly of the G0/G1 phase as that of the S phase and G2/M phase, and the cells were locked in S phase. After 48 h when treating drugs, the number of mitochondria was increased and the appearances were different in all-trans retinotic acid group, and double membrane and endocristae were clear. In sodium selenite group, the outline and framework of mitochondria were vague and some mitochondria looked swollen. In the combination group of two drugs, the endocristae of mitochondria looked shortened and decreased, some atypic masses of high electronic density were found. CONCLUSIONS: All-trans retinotic acid which is lipophilic can combine with mitochondria and reinforce its function. Sodium selenite which is hydrosoluble is uneasy to combine with mitochondria, which inhibites the function of mitochondria. It is deserved to study more that two drugs effect on mitochondria of tumor cells.
