Effect of Intrathecal Nefopam on Cyclooxygenase-2 Expression in Dorsal Horn of Spinal Cord in Rat with Peripheral Pain

Abstract: Objective To investigate the effect of intrathecal(IT) nefopam on the expression of cyclooxygenase-2 in dorsal horn of spinal cord in rat model with peripheral pain.Methods Thirty-two male SD rats were randomly divided into 4 groups(n=10 each):GroupⅠ(100 μL normal saline was injected into the plantar surface of left hindpaw.).Peripheral pain was induced by injection of 5%formalin 100 μL into the plantar surface of left hindpaw in GroupⅡ,group Ⅲ and group Ⅳ.In group Ⅲ,normal saline 20 μL was injected intrathecally before interplanetary formalin injection.Nefopam 5 μL+ normal saline 15 μL was injected in group IV.Pain behavior was assessed by PIS score.All the animals were killed at 2 h after interplanetary formalin (or NS) injection after assessment of pain behavior and cyclooxygenase-2 expression was determined.Results The cyclooxygenase-2 expression in group Ⅱ and Ⅲ was significantly higher than that in group I but it was significantly lower in group Ⅳ(P<0.05).Conclusion Intrathecal nefopam could inhibit the cyclooxygenase-2 expression in the spinal dorsal horn induced by interplanetary injection of formalin.
