The Role of P-selectin in the Pathogenesis of Diabetic Nephropathy

Abstract: P-selectin is a member of adhesion molecular family. It mediates platelet-leukocyte interaction as well as endothelium-leukocyte adhesion in inflammatory and thrombotic reaction. P-selectin could be considered as a disease marker in wide-spread disorders including artherosclerosis, malignancy, venous thrombosis and kidney diseases, etc. In diabetic nephropathy an increase in P-selectin was detected both in the kidney tissues and circulating blood. Hyperglycemia promotes leukocytes adhesion to the intraglomerular endothelial and activated platelets through upregulation of cell surface expression of P-selectin, which is possibly depended on NF-κB activation and PKC cell signal pathway. The infiltration of leukocytes into the glomeruli and interstitium play an important part in the progress of diabetic glomerulosclerosis and interstitial fibrosis.
