Expression of Osteopontin in Proliferated Human Umbilical Artery Smooth Muscle Cell Induced by Endothelin-1

Abstract: Objective To study the variation of the expression of Osteopontin (OPN) in the proliferated human umbilical artery smooth muscle cell (HUASMC) induced by ET-1.Methods ①HUASMCs were cultured by explant method;②Cultured HUASMCs were divided into 5 groups randomly:Group 1, group 2, group 3 (treated with 100 nmol/L,10nmol/L,and 1nmol/L ET-1 respectively), group 4 without treatment by ET-1, and group 5 (pretreated with 10μmol/L BQ123 and then cultured in 10nmol/L ET-1). All of the cells were cultured for 48 hours. The level of OD490 was evaluated in different groups by MTT assay;③The expression of OPN was measured by immunocytology, and the positive rate was compared among groups;④The relationship between the proliferation of HUASMC and the expression of OPN was analyzed.Results The higher the concentration of ET-1, the higher of OD490, and the differences were significant (P<0.01).ET-1 increased the expression of OPN dose-dependently (P<0.01).The expression of OPN was correlated with the proliferation of VSMC(r=0.962,P<0.01).Conclusion ET-1 could promote the proliferation of VSMC dose-dependently.OPN expression increased significantly after treated with ET-1.OPN expression was correlated with VSMC proliferation positively.The examination of OPN might indicate the early proliferation of VSMC.
