Effects of YTT on mRNA Expression of C-myc and PDGF-A in the Patients with IRS

Abstract: Objective:To explore the molecular mechanism of Yangxin Tongmai Tablet (YTT) in preventive and curative insulin resistance syndrome (IRS) through checking the effects of YTT on mRNA of C-myc and platelet derived growth factor-A (PDGF-A) in unit-nuclear cell of peripheral circulation. Methods:77 cases with IRS patients were randomly divided into 2 groups, viz. 33 and 44 cases treated with YTT and metformin (met), respectively. Fasting plasma glucose (FPG) and insulin(FINS), etc. were measured by the conventional method, while the expression level of mRNA of C-myc and PDGF-A was tested with the means of reverse transcriptase-polymerase chain reaction (RT-PCR). Results:The contents of FPG and FINS after treatment showed significantly dropper (P<0.01), while insulin sensitivity index (ISI) was higher and the expression level of mRNA of C-myc and PDGF-A was lower than before treatment (P<0.01). However, compared with met group, the level of c-myc mRNA expression in YTT group was remarkably reduced (P<0.01). Conclusion:YTT had the roles with inhibiting over proliferation of vascular smooth muscle cells and preventing formation and development of IRS through improving insulin sensitivity, inhibiting C-myc mRNA expression and following reducing the level of PDGF-A mRNA.
