Study on Extraction of Total Alkaloid from Konjac Powder

Abstract: Objective:To Optimize the alkaloid extraction procedure of total alkaloid from Konjac Powder.Methods:The total al- kaloids were measured at 414nm with the spectroscopy by means of acid dye method.Six factors were tested in single factor experiment. These six factors included the concentration of alcohol,the ratio of sample to alcohol,pH,temperature,time and withdraw times,which could influence the yield of total alkaloid.The alkaloid extraction procedure was optimized by the orthogonal experiment after single fac- tor experiment.Results:The results indicated that four factors influenced the extraction efficiency:pH value>alcohol concentration>ra- tio of sample to alcohol>temperature.The pH values is of significance(P<0.05).The optimum condition was that the ratio of sample to alcohol was 1:10,the temperature of extraction was 50℃,alcohol concentration was 70 % and the pH was 1.Under this condition,the content of the alkaloid from Konjac Powder was 89.79 mg/100g.Conclusion:This technology is simple and feasible.It provides a theo- retical basis for extracting the total alkaloid of Konjac Powder.
