Abstract: Objective To determine survivin(SVV)variant in human hepatoma cell line-HepG2 and to construct its eukaryotic expression vector.Methods The total RNA of HepG2 was extracted;3 special primers were designed and identified;SVV-3 primers containing double restriction cndonuclease BamHⅠand XhoⅠwere designed;the whole coding area of SVV-3 was amplified by RT-PCR;the amplified product was double-digested by BamHⅠand XhoⅠand cloned into pcDNA3.1,detecting its sequence.Results SVV-3 was expressed in HepG2 more than SVV-1.The sequence is consistent with SVV-3 sequence published in Gen-Bank. Conclusions SVV-1 and SVV-3 were detected to be expressed in HepG2.A recombinant plasmid vector containing human SVV-3 was constructed successfully.
