Abstract: Objective To investigate the effect of one-month intermittent levodopa(L-dopa) administration on behaviour in rats with Parkinsonism and establish the rat model of L-dopa induced dyskinesias(LID).Methods Unilateral 6-OHDA lesioned rat model of Parkinson's disease(PD) was developed by injecting 6-OHDA in the Medial forebrain bundle(MFB).Those successful PD rats were divided into two groups(A&B).The rat received L-dopa(A) or saline(B) injection once daily for 1 month,respectively.The behaviour change was observed,abnormal involuntary movement(AIM)was written down and apomorphine induced rotation was counted.Results Most rats were proved to be successful by apomophine inducting.The ratio is 75%.Rats with PD had behaviour change in the treatment,the rotation counts increased from the first 2 to 3 weeks and then decreased gradually.In A group 72% of them developed AIM.Conclusions Through MFB point injection the rat model in PD is developed in high ratio.Rats with PD have behaviour characterization in our observation.The pulsatile L-dopa administration and the severity in rats may be involved in the formation of LID.
