Abstract: Objective To seek an ideal material for urethral reconstruction.Methods Human amnion extraeellular matrix (HAECM) was obtained from health fresh human amniontic membrane by decellularization,whose effect was observed under the inverted microscope.In the experimental group of 9 male rabbits,a segment of the urethra with the length of 1cm was resected and replaced by the HAECM with equal length and calibration.Sham operation was carried out in the control group of 3 rabbits.The urethra was exposed and carefully examined before the sacrifice of rabbits at 4,12 and 16 weeks.The whole specimen was then excised for histopathological examination.Results All rabbits survived after surgery before sacrifice.In the experimental group,all the HAECM remained in its original position,of which one had infection complicated with fistula and 1 had simplex fistula.In the control group,all of the rabbits were normal.Histopathological examination of the experimental group showed there were gradual regeneration over the HAECM and the continuity of the new urethra recovered at 4 weeks.Sporadic smooth muscle bundles and slight regenerated vessels could be observed at 12 weeks.Sixteen weeks after the operation,there were more capillary vessels and smooth muscles in the new urethra,but compared with the control,the array of smooth muscles was still in disorder.Conclusions HAECM tube is able to repair the defect of the rabbit urethra.
