Alterations in MAPK signaling pathway in human airway epithelial injury

Abstract: 目的:利用基因芯片技术研究多聚左旋精氨酸(PLA)损伤时人气道上皮细胞丝裂原活化蛋白激酶(MAPKs)信号通路相关基因的表达变化.方法:将培养的人气道上皮细胞株16HBE分为对照组和PLA损伤组(终浓度8 μmol/L),PLA处理10 min后分别收集细胞,抽提总RNA.运用基因芯片及生物信息分析技术检测分析MAPK信号通路中96种相关信号分子的表达.结果:PLA损伤组与对照组比较,MAPKs通路异常激活,Cyclin D1、p38 MAPK、ERK2、JNK1等相关信号分子表达上调.结论:气道上皮细胞损伤时MAPKs信号通路的异常激活.参与了上皮细胞合成、释放多种细胞因子和趋化因子的过程.
