Abstract: 目的:获得高纯度的Ⅱ型胶原,并考察其对类风湿性关节炎的作用;方法:以鸡胸软骨为原料制备Ⅱ型胶原,在酸性条件下添加胃蛋白酶酶解鸡胸软骨Ⅱ型胶原三股螺旋链的端肽,采用DEAE-sepharose CL 6B离子交换柱纯化酶解样品制得Ⅱ型胶原;通过SDS-PAGE电泳、氨基酸分析及X-射线衍射表征Ⅱ型胶原的纯度与结构;建立大鼠类风湿性关节炎模型(CIA),研究不同浓度的Ⅱ型胶原对大鼠类风湿性关节炎的影响;结果:经过纯化的Ⅱ型胶原纯度为95.21%.呈典型的三股螺旋结构;口服Ⅱ型胶原能明显降低CIA大鼠的关节炎指数,缓解关节红肿等症状;同时,实验组大鼠的T淋巴细胞增殖及白细胞介素-2(IL-2)的分泌受到抑制,白细胞介素-1(IL-1)及肿瘤坏死因子-α(TNF-α)水平也显著低于阴性对照组.结论:Ⅱ型胶原能作为膳食补充荆防治人类类风湿性关节炎.
