THE EFFECT OF LOW-DOSE CISPLATIN COMBINED WITH INTERFERON-ALPHA ON ANGIOGENESIS IN MICE WITH TRANSPLANTATION LIVER CANCER

Abstract: 目的 了解持续低剂量顺铂(DDP)联合α-干扰素(IFN-α)对小鼠移植性肝癌血管生成的影响.方法 将48只接种H22肝癌的昆明小鼠随机分为4组,各12只. 低剂量DDP组(DDP组):腹腔注射DDP 0.2 mL(0.6 mg/kg);IFN-α组(IFN组):皮下注射IFN-α 0.2 mL (2×104 U);低剂量DDP联合IFN-α组(联合组):腹腔注射DDP 0.2 mL (0.6 mg/kg)并皮下注射IFN-α 0.2 mL (2×104 U);生理盐水对照组(对照组):腹腔注射生理盐水0.2 mL,每天1次,连续2周.观察肿瘤体积、小鼠体质量的变化和药物毒副反应,采用免疫组织化学方法测定肿瘤微血管密度(MVD)、血管内皮生长因子(VEGF)的表达.结果 DDP组和IFN组肿瘤生长比较缓慢,联合组抑瘤效果更为明显(F=260.58,q=0.72~1.08,P<0.05),且毒、副反应未增加.与对照组比较,DDP组和INF组的MVD、VEGF表达均下降(F=157.04、 52.97,q=9.55~19.02,P<0.05),联合组下降更显著(q=5.25~9.47,P<0.05).结论 低剂量DDP与IFN-α联合应用具有明显的抗血管生成协同作用,抑瘤效果显著,毒、副反应小.
