Abstract: 目的 研究急性暴露尼古丁(nicotine)对未成年大鼠脑内神经细胞凋亡的影响.方法 用免疫组织化学法和Western blotting法检测急性暴露尼古丁的未成年模型大鼠脑内Caspase-3的表达;用原位末端标记法(TUNEL法)检测凋亡细胞.结果 免疫组织化学法检测到大脑皮层、海马齿状回、CA2区和CA3区的Caspase-3表达在实验组明显增高.蛋白质免疫印迹法(Western blotting)检测到大脑皮层和海马Caspase-3原型及活性片段的表达在实验组明显增高.TUNEL法检测到大脑皮层和海马齿状回的阳性细胞在实验组明显增多.结论 急性暴露尼古丁激活未成年大鼠脑内大脑皮层和海马内神经细胞Caspase-3,促进大脑皮层和海马齿状回的神经元凋亡.
